Participates in projects in which protein sequence variants of drug candidates are generated. Perform protein purification, Protein Characterization and sample management. Promising drug candidates are expressed, purified and examined for receptor binding as well as for effectiveness in cell-based assays and in animal disease models. Responsibilities include the following: Purifying antibodies and other proteins (Protein A, SCX, Ni-NTA) using automated protein purification and fractionation systems (AKTA); characterizing proteins by Size Exclusion Chromatography (SEC); performing protein concentration and buffer exchange using Tangential Flow Filtration and centrifugal spin columns; managing and organizing inventory of recombinant proteins; managing and overseeing sample distribution and shipment; generating DNA constructs for protein expression in mammalian cells (Restriction enzyme-assisted cloning, Gibson Assembly, PCR mutagenesis and sub-cloning into expression vectors); measuring Endotoxin levels in protein; analyzing Proteins by SDS-PAGE and immunoblotting; and inventory management of laboratory reagents and maintaining experimental procedures in electronic lab notebook.
Must have MS in biotechnology or related field and 2 years of industry experience generating DNA constructs for protein expression in mammalian cells through restriction enzyme-assisted cloning, Gibson Assembly, PCR mutagenesis and sub-cloning into expression vectors. Experience must include analytical lab work; Unicorn protein purification software (or equivalent software); and Snapgene and/or Geneious sequence analysis software. We will also accept a BS and 5 years of experience.
